What it is
Andarine (also called S-4) is a synthetic, nonsteroidal selective androgen receptor modulator (SARM) originally developed by GTx, Inc. to treat conditions such as muscle wasting, osteoporosis and benign prostatic hyperplasia.[1][3] It was never approved for human use and is sold only as a "research chemical"; in fitness communities it is used mainly as a "cutting" compound for lean mass, fat loss and muscle hardness.[2][6]
How it works
Andarine binds with high affinity to the androgen receptor and acts as a partial/tissue-selective agonist, stimulating anabolic signaling in skeletal muscle and bone while producing fewer androgenic effects in tissues like the prostate than testosterone or anabolic steroids.[1][4] At the cellular level it activates anabolic pathways (e.g. mTOR signaling) to increase muscle protein synthesis.[5] It is orally bioavailable, so no injection is needed.[2] Notably, andarine also binds receptors in the eye/retina, which is thought to underlie its characteristic visual side effects.[8]
Risks & side effects
Most important: Andarine's most distinctive risk is dose-dependent vision disturbance: users frequently report a yellow tint to vision, impaired night vision and difficulty adjusting between dark and light, typically emerging around 50 mg/day or higher.[7][6] These effects are reported to be reversible after stopping, but more broadly SARMs are unapproved drugs that the FDA links to life-threatening reactions including liver injury requiring hospitalization.[6][9]
